Abstract
Purpose: This work was performed to produce the therapeutic monoclonal antibody (X01 Mab) against a clinically validated Epidermal Growth Factor (EGF) receptor.

Materials and Methods : X01, monoclonal antibody against EGFR receptor, was developed through Phase display technology with immune library. In order to analyze the characteristics of the developed antibody and its in vitro activity in the cell level (proliferation inhibition), SEC-HPLC and SDA-PAGE experiments were conducted. In addition, the efficacy of X01 MAb in vivo was also confirmed by checking the eradication of A431 tumor cell mouse xenograft. And to understand
its mechanism of action, effects of X01 MAb in the activity of cell signaling molecules involved in EGF signaling in A431 cells were conformed. For developments of measuring the contents of X01 MAB in the biological sample (serum / plasma), recombinant proteins against the extracellular domain (ECD) of EGFR were manufactured and purified.

Result: X01, a monoclonal antibody specifically binding to EGFR was developed. X01 MAb was capable of killing A431 tumor cells which was confirmed in A431-used Xenograft mouse model. This antibody¡¯s capacity was believed to be similar to the efficacy of Cetuximab currently available for the treatment of colon and head & neck cancers, showing the possibility that the antibody can be developed further as a therapeutic monoclonal antibody. X01 MAb was confirmed to block both the phosphorylations of EFGR and Erk, a signal transducer in EGF signaling pathway.

Conclusion: A monoclonal antibody against a clinically evaluated EGF receptor was developed in order to check whether the antibody can be developed further as a therapeutic monoclonal antibody
